The blaze took hold at St Coleman's Park, Macroom at about 4 o'clock this morning.
A woman who jumped from an upstairs window from the property and a male neighbour who tried to get into the house to help – have both been brought to hospital with non life threatening injuries.
Post mortem examinations on the men – one in his 60's and the other in his 20's – are due to be carried out at Cork University Hospital.
Gardaí say the fire appears to have started accidentally.
