Mullah Akhtar Mansour was targeted along with another militant in Pakistan near the Afghan border.
A senior Taliban official confirmed that Mansour died in the attack.
A US official, speaking on the condition of anonymity, says multiple drones were aimed at the men as they rode in a vehicle in a remote area.
The strike is reported to have been authorised by President Barack Obama.
